Triple Spectrometer

A triple quadrupole mass spectrometer (TQMS) is a tandem mass spectrometer with three quadrupoles arranged in series, enabling highly sensitive and selective analysis of complex samples.

Overview and Design

A triple quadrupole mass spectrometer consists of three quadrupoles in series, commonly denoted as Q1–q–Q3. Q1 and Q3 are mass-resolving quadrupoles that filter ions based on their mass-to-charge ratio (m/z), while the middle quadrupole, q, acts as a collision cell where ions undergo collision-induced dissociation (CID) without mass resolution . This configuration allows the instrument to perform tandem mass spectrometry (MS/MS), providing structural information and highly selective detection of target compounds .

Operating Principles

  • Q1 (first quadrupole): Selects precursor ions of interest from the ion source.
  • q (collision cell): Ions collide with an inert gas (e.g., argon), fragmenting into product ions.
  • Q3 (third quadrupole): Filters the resulting product ions for detection. This setup enables selected reaction monitoring (SRM) or multiple reaction monitoring (MRM), which are highly sensitive and specific modes ideal for quantitative analysis of trace compounds .

Applications

Triple quadrupole mass spectrometers are widely used in:

  • Pharmaceutical and clinical research: Quantifying drugs, metabolites, and biomarkers in complex biological matrices .
  • Environmental analysis: Detecting pesticides, PFAS, and other contaminants at ultra-trace levels .
  • Food safety testing: Measuring mycotoxins, steroids, and other residues with high precision .
  • Biopharmaceutical quality control: Ensuring reproducibility and compliance in high-throughput laboratories .

Advantages

  • High sensitivity and selectivity: MRM allows detection of compounds at very low concentrations.
  • Robustness in complex matrices: Modern instruments like the SCIEX 7500+ and Thermo TSQ Plus maintain sensitivity even in challenging samples .
  • Versatility: Can operate in full-scan, selected ion monitoring, or MRM modes depending on analytical needs .
  • Efficiency: Advanced ion optics and collision cells improve throughput and reduce maintenance downtime .

Modern Instruments

Examples of current triple quadrupole systems include:

  • Thermo Scientific TSQ Certis and TSQ Plus: Designed for high-throughput, compliance-ready workflows with enhanced sensitivity and reproducibility .
  • SCIEX Triple Quad 7500+: Offers ultrafast MRM acquisition, high sensitivity, and resilience in complex matrices, suitable for lipidomics, PFAS, and mycotoxin analysis . Triple quadrupole mass spectrometers remain a gold standard in quantitative mass spectrometry, combining high resolution, selectivity, and sensitivity for a wide range of analytical applications.

Triple quadrupole mass spectrometer

A triple quadrupole mass spectrometer (TQMS), is a tandem mass spectrometer consisting of two quadrupole mass analyzers in series, with a (non-mass-resolving) radio frequency (RF)–only quadrupole between them to act as a cell for collision-induced dissociation. This configuration is often abbreviated QqQ, here Q1q2Q3.
